Distinct microbial mediators link diet to inflammation in Crohn’s disease and ulcerative colitis: This longitudinal study demonstrated that diet quality influences intestinal inflammation through distinct microbiome pathways. Specific taxa and metabolites mediated effects in Crohn’s disease, whereas microbial richness and global composition drive protection in ulcerative colitis.

No effect of computer-aided diagnosis on colonoscopic adenoma detection (CADe): CADe did not increase the adenoma detection rate (ADR) in a large pragmatic multicenter randomized study involving 1627 patients older than 50 years with mixed colonoscopy indications. ADR was significantly influenced by the examiner, patient age and sex, while more hyperplastic polyps were found by CADe.

Whole-exome sequencing in unexplained adult-onset cholestatic liver disease: In this single-center study, whole-exome sequencing (WES) was performed in 21 adults with unexplained cholestatic liver disease. A genetic diagnosis was established in 5 patients (23.8%), with 3 of the 5 identified cases involving genes outside the classical PFIC spectrum. These findings suggest that WES may be a valuable diagnostic tool in the evaluation of patients with unexplained cholestatic liver disease.

Prevalence of liver fibrosis in the European general population: In the multinational European LiverScreen study, including 30,199 participants, the prevalence of liver fibrosis in the general population was assessed using non-invasive screening methods. Increased liver stiffness was detected in 4.6% of individuals, and approximately one-third of these were subsequently confirmed to have chronic progressive liver disease. Steatotic liver disease was by far the most common underlying etiology. These findings support targeted screening strategies for the early detection of advanced liver disease.

Camrelizumab plus CAPOX (capecitabine plus oxaliplatin) vs. CAPOX alone as initial treatment for gastric or gastro-esophageal junction adenocarcinoma: First-line therapy with the PD-1 inhibitor camrelizumab plus CAPOX followed by camrelizumab based maintenance was associated with longer overall survival than CAPOX in HER-2 negative unresectable locally advanced or metastatic cancer in this Chinese phase 3 trial.

Perturbations of diet and gut signatures persist during remission in Crohn’s disease (CD) despite effective immunosuppression: Analysis of diet, microbiome metabolomics, ileal transcriptomics and clinical data revealed that signature perturbations associated with active inflammation also persist during remission. These findings underline a potential explanation for the remitting-relapsing nature of CD and suggest that interventions targeting these parameters may promote deeper remission states.

No prevention of recurrent pancreatitis by simvastatin: A randomized trial evaluating simvastatin vs. placebo in patients with recurrent acute or chronic pancreatitis did not demonstrate a reduction in pancreatitis recurrence rate. Interestingly, neo-onset diabetes was observed more frequently in simvastatin-treated patients.
